Duncan Howie
Director, Cell & Immunobiology Enara Bio
Duncan is Director of Cell & Immunobiology at Enara Bio, where he leads a team focused on the discovery and early development of T cell engager therapies for solid tumours. His role spans target selection, assay development, and format optimisation, with a focus on translating biological understanding into candidates suitable for preclinical progression. He has contributed to the development of Enara’s approach to targeting tumour-restricted antigens and to evaluating effective T cell engager formats.

Panel Discussion: Cracking the ‘Clean Target’ Challenge with Smarter Selection Strategies to Uncover Safe & Novel Targets
11:00 am
- What defines a ‘clean’ target in practice, and how should we balance prevalence, selectivity, and heterogeneity risks?
- How can emerging technologies like proteomics, spatial biology, and AI improve prediction of protein expression, localisation, and off-tumour toxicity risk?
- How can the field move beyond a limited pool of validated targets while generating robust preclinical data to de-risk novel target investment?

Dancing in the Dark: Discovery, Validation, & Clinical Application of Dark Antigens®
9:30 am
- Unlocking a new target class: The “Dark Genome” is a rich, untapped source of tumour-specific antigens, overcoming the limitations of many conventional TCE targets (poor tumour specificity, low prevalence, low intra-tumoural homogeneity)
- End-to-end platform solution: Enara Bio has built integrated discovery (EDAPT) and engineering (EnTiCE) platforms to identify Dark Antigens® and generate optimised TCEs capable of effective engagement at physiological antigen levels
- First-in-class validation with DARKFOX/ENA101: We have identified a highly tumour-specific, broadly expressed antigen and targeted it with a potent, selective TCE showing strong preclinical efficacy, safety, and translational potential
